The Molecular Probes™ BioParticles™ product line includes a line of fluorescently labeled thermally or chemically inactivated bacteria and yeasts in sizes, They vary in shape and natural antigenicity. These fluorescent BioParticles™ products have been used to study phagocytosis by fluorescence microscopy, quantitative spectrometry and flow cytometry.

Staphylococcus aureus (Wood strain, no protein A) and Zymosan (
Saccharomyces cerevisiae) BioParticles™ conjugates are covalently labeled with a variety of different fluorophores (special care has been taken to remove free dye after conjugation). Unlike the fluorescence of fluorescein-labeled BioParticles™ conjugates, which is partially quenched in acidic environments, the fluorescence of Alexa Fluor™, BODIPY™ FL, tetramethylrhodamine, and Texas Red™ dye conjugates Uniformly bright in the pH range 3 to 10.

E. coli or
Staphylococcus aureus particles per mg solids, and approximately 2 x 107 zymosan particles per mg solids. BioParticles™ conjugates can be reconstituted in the buffer of choice for phagocytosis assays. The fluorescence of BioParticles™ conjugates bound to the cell surface (but not endocytosed) can be quenched by ethidium bromide, trypan blue, or other quenching groups. In addition to cell applications, fluorescent BioParticles™ conjugates can also be used in flow cytometry calibration reference materials for sorting bacterial and yeast mutant strains. These small particles may also be useful references for light scattering studies because they vary in size and shape.
